Abstract:

The aim of these research was to analyze the differences between 10 species Selaginella by DNA-AFLP method. The research consisted of (i) selection of AFLP primer which can produce polymorphic bands, and (ii) genetic similarities analysis, UPGMA analysis, and principal component analysis (PCoA). DNA was isolated from c. 0.5 g silica dried leaves. Total genomic DNA was extracted using CTAB procedure. PstI/MseI used to digest DNA, while selective amplification primes was P-11 plus either M-48, M-48, M-50, or M-51. The results show that 4 primer combinations at 10 species Selaginella produced 248 polymorphic bands, in the range of 1-56. Selaginella alligans, Selaginella opaca, Selaginella cf. plana, and Selaginella caulescens had good polymorphic bands because could generate about 30-60 bands, while the other only generate < 20 bands. Generally, AFLPDNA method could generate fingerprinting to differ Selaginella species, whereas genetic similarity, UPGMA, and PCoA analysis showed that each species had diverse similarity index, in the range of 59-100%. But, some species generate little polymorphic bands while the other generate flourish bands. It was need to search selective primer combination that could be used as general primer.
