Abstract:
Until now mistletow hesitation at teak
plantation in Jawa in Perhutani region has extended in a
widespread scale of areal extents and distribution in teak plots in Forest
authority units (FAU/KPH) and Clonal seed garden (CSO), especially CSO
Padangan. To support early detection system for controlling teak mistletoe is
needed supporting basic research among others ‘Diversity analysis and DNA
barcode as reference for applied research. Purpose this research is DNA Analysis for parasitic
plant (mistletoe) diversity on a stand of teak, and apply DNA Barcode technical
with nukleotida difference analysis and genetic distance to determine mistletoe
species hesitate teak stand. Research procedure for diversity DNA covering take
and treat sample, extraction and testing DNA quality, process PCR-RAPD, data
analysis usage POPGENE versi 32 dan NTSys. For DNA barcode covering
identify teak mistletoe species based of morphology, take leaves samples for
measuring leaf morphologi character and DNA Barcode covering PCR with core
marker (matK, rbcL), sequencing DNA, documentation, DNA Barcode analysis and
filogenetik. By multivariate analysis (PCA, MCA) can distinguish two teak
mistletoe Dendrophthoe pentandra and M.
tetragonus. Reinforced result significant different test for every leaf
morphology variable those mistletoe showed some variable different significant i.e petiole length,
leaf width, sum secondary vein, sum leaf, leaf base form, aspect ratio, form factor, perimeter ratio. Besides, primer matK dan rbcL that used at DNA Barcode
can distinguish based filogenetic among
D. pentandra, M. tetragonus and Viscum articulatum so two core primer were recommended usage for
DNA Barcode teak mistletoe. As for deteminating genetic diversity of mistletoe
teak, exactly marker RAPD (OPP 9, OPBH 16, OPBH 20), can amplification success
to teak mistletoe of D. pentandra.
