Abstract:
Primary selection is an effective method to develop SSR
markers showing close kinship of several species in a taxonomic system.
This study was aimed to measure diversity genetic of sengon
resistant dan suceptible to boktor pest and gall rust disease based on
microsatelit marker. Leaves samples of resistant and susceptible sengon to gall rust disease were collected from Kediri, and Sukabumi,
while Leaves samples of resistant and susceptible sengon to boktor pest from Sumedang. Amplifications of eight SSR markers (Pafa02, Pafa03, Pafa04, Pafa05,
Pafa06, Pafa07, Pafa08, and Pafa10) were tested on the sengon resistant and susceptible to gall rust from Sukabumi and Kediri and four SSR markers
(Pafa06, Pafa07, Pafa08 dan Pafa10) were tested on the sengon
resistant and susceptible to boktor from Sumedang. Results of analysis
showed informative markers,
i.e. average value of
He = 0.479 to gall rust ; He 0.637
to boktor,
respectively. The SSR markers
could amplify most of sengon accessions used in this study. Molecular analysis showed high He value indicating the high
genetic diversity of populations studied.
The markers used could distinguish sengon accessions which were
resistant to gall rust disease from the susceptible.
