Screening and Isolation Isolation of Bioactive Compounds to control Hypertension

Abstract:

The present study was undertaken to find the antihypertensive value of 23 traditional herbal medicines through the screening based  on their ability to scavenge the stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical to detect active principle.  Antioxidants have been reported to prevent oxidative damage caused by free radical and can be used in cardiovascular diseases. The phytochemical analysis of 23 species of herb revealed the content of alkaloid, saponin, tannin, steroid/ terpenoid and flavanoid class of compounds of those species.  The radical scavenging activity tests of the methanolic crude extracts of all spcies has been studied. The high radical scavenging activity was observed  in the leaves of Pilea trivernia followed by leaves of Phyllanthus urinarian, Andrographis paniculata and Piper betle. They displayed antioxidant activity. An activity-directed fractionation and puri??????cation process was used to identify the DPPH (l,l-diphenyl-2-picrylhydrazyl) free radical-scavenging components of Pilea trivernia.  The powder Air dried leaves of P.trivernia were extracted with methanol. The methanolic crude extract was reextracted with hexane. The hexane phase showed strong DPPH radical-scavenging activity  was then subjected to separation and puri??????cation using preparative thin layer chromatography using silica gel.  The compound indicated strong DPPH radical-scavenging activity was isolated and analysis of the compound with GC-MS is under way to identify the chemical structure as well as the hypotensive activity by bioassay tests with rat.
